EFFECT OF PIOGLITAZONE AND IRBESARTAN ON HEMODYNAMIC AND METABOLIC PARAMETERS IN FRUCTOSE INDUCED DIABETES AS WELL AS HYPERTENSIVE RATS

Abstract

Combination of Pioglitazone (AT1 blocker) and Irbesartan (PPAR-γ agonist) is the most drug therapy used in diabetes hypertension. The aim of this study is to the effects of Irbesartan and pioglitazone alone and in combination, in the fructose overloaded rat, a model of diabetes hypertension. Fructose feeding caused significant elevations in blood pressure, blood glucose and the concentrations of plasma triglycerides and free fatty acids. High-density lipoprotein cholesterol (HDL-C) was significantly reduced and very low-density lipoprotein cholesterol (VLDL-C) was significantly elevated. All six week treatments groups reduced blood pressure, blood glucose triglyceridemia, total cholesterol and increased level of HDL and VLDL clearance. In conclusion, combination therapy significantly produced additive effect rather than single drug on hemodynamic and metabolic changes.
